目的:开发冻干人用狂犬病疫苗(鸡胚成纤维细胞)的连续流蔗糖密度梯度离心纯化工艺。方法:分别比较两种不同初始蔗糖浓度和不同上样速度对纯化效果的影响,初步确定纯化工艺;通过多批次实验确定样品收集范围;比较不同浓缩倍数条件下杂质去除和抗原回收情况,确定合适的收获液浓缩比例;比较不同批次样品纯化后的杂质去除率和重复性,判定本纯化工艺的稳定性。结果:选取60%作为初始蔗糖浓度,在上样速度为150~200ml/min时,可以有效地对10倍浓缩的病毒收获液进行纯化;卵清蛋白、牛血清白蛋白和庆大霉素去除率分别达到99%,95%和95%,且工艺具有极好的稳定性。结论:开发的连续流蔗糖密度梯度离心技术可以作为冻干人用狂犬病疫苗(鸡胚成纤维细胞)的产业化纯化工艺。  相似文献

本实验建立一种快速从灵芝子实体中制备灵芝烯酸B的方法。以沪农灵芝1号子实体乙醇提取物为原料,经过D101大孔树脂粗分,用60%乙醇洗脱后,采用高速逆流色谱对获得的灵芝酸性三萜进行分离制备。确定最佳溶剂体系为正己烷-乙酸乙酯-甲醇-水(V/V/V/V,5:5:2:9),上相作为固定相,下相作为流动相,单因素法和正交实验设计确定高速逆流色谱法分离灵芝烯酸B的最佳工艺为:流速2.0mL/min,转速900r/min,一次上样量为500mg时,制备得到灵芝烯酸B化合物得率为(9.07±0.16)%,纯度为(85.04±3.45)%。用质谱、核磁对得到的灵芝烯酸B进行了结构鉴定。此法操作简单高效,为大量制备灵芝烯酸B提供了参考。  相似文献

Where data and information are lacking, structured expert risk assessments can provide a powerful tool to progress natural resource planning. In many situations, practitioners make informal assessments of risk within small groups that typically constitute employees. In this study, we report on three small (in terms of experts) structured expert‐based risk assessment case studies conducted by expert employees of a not‐for‐profit organisation (Australian Wildlife Conservancy) to demonstrate the utility of the approach. The case studies were carried out for three wildlife sanctuaries managed by AWC: Faure Island, Karakamia and Paruna. The likelihood that a set of direct risk factors would cause management failure for sets of important wildlife elements in the three sanctuaries was elicited from the small group of ecological experts. The analysis was couched in terms of a management aim to not lose species from each wildlife element over the management period of 25 years with current management. The experts believed, in particular, that increasing temperature and decreasing water availability associated with climate change were likely to impact significantly upon the vegetation elements and water‐reliant fauna associated with the sanctuaries. Some vegetation elements were also thought likely to be at risk of over‐grazing, unsuitable fire regimes and, in some cases, disease. In addition to predation by exotic predators at one sanctuary, the experts identified additional direct risk factors for various fauna elements associated with expected changes to the vegetation elements, including reduction in food availability, nesting habitat, and generally important life media. From the risk analyses, a preliminary conceptual model was developed to underpin monitoring and to indicate areas for possible management intervention and research. The case studies demonstrate that even in a small workplace team, structured risk assessments can be efficiently accomplished and can provide expedient and transparent information that effectively captures and aggregates the views of the experts.  相似文献

以紫斑牡丹(Paeonia suffruticosa var. papaveracea)花瓣为原料,采用隔离窨制,对牡丹花茶窨制过程中花坯、配花量等主要影响因子进行研究。结果表明,密闭箱桶温度21 ℃、相对湿度90%、窨制时间48 h条件下经一窨一提获得的花茶,感官评审花与茶叶的协调度高、香气高锐持久、茶汤滋味醇正鲜爽。采用气相色谱-质谱联用仪分析花茶香气成分,新鲜花瓣与茶叶配比5:1窨制48 h的花茶苯乙醇、香叶醇、橙花醇含量较高,其香气高扬、茶汤醇正鲜爽,配比2.5:1的花茶上述成分含量和感官其次,而拌和型茶品透素欠鲜爽。发酵或发酵揉捻花瓣窨制的花茶乙醇、环氧芳樟醇及高级烷烃含量较高,其主要赋香物质低于新鲜花瓣含量,渥味明显,茶汤有浊气欠鲜爽。  相似文献

Claimants and industry professionals frequently view conflict that arises in the course of a native title claim as a detriment to timely claims resolution. I argue instead that disputation itself may constitute an integrative social process through which participants define, delimit and reproduce community. I show also how ethnographic analysis of disputation can provide useful insights into broader social and cultural practices and normative value systems. Drawing on theoretical and methodological concerns with human agency and the integrative and constitutive role of conflict that challenges consensus models of community, I develop a practice‐centred approach that is attentive to the ways that participants in a dispute articulate and contest the definition of community. The possibility of viewing 'culture' and group cohesion as contingent and emergent through disputation should assist anthropologists working in the native title field to incorporate conflict as a productive aspect of social and cultural practice.  相似文献

Accurate species delimitation has a pivotal role in conservation biology, and it is especially important for threatened species where decisions have political and economic consequences. Finding and applying appropriate character sets and analytical tools to resolve interspecific relationships remains challenging in lichenized fungi. The main aim of our study was to re-assess the species boundaries between Usnea subfloridana and Usnea florida, which have been phylogenetically indistinguishable until now, but are different in reproductive mode and ecological preferences, using fungal-specific simple sequence repeats (SSR), i.e. microsatellite markers. Bayesian clustering analysis, discriminant analysis of principal components (DAPC), minimal spanning network (MSN), and principal component analysis (PCA) failed to separate U. florida and U. subfloridana populations. However, a low significant differentiation between the two taxa was observed across all populations according to AMOVA results. Also, analysis of shared haplotypes and statistical difference in clonal diversity (M) supported the present-day isolation between the apotheciate U. florida and predominantly sorediate U. subfloridana. Our results do not provide a clear support either for the separation of species in this pair or the synonymization of U. florida and U. subfloridana. We suggest that genome-wide data could help resolve the taxonomic question in this species pair.  相似文献

人类活动排放大量的CO2通过海气界面进入海洋,打破原有海水碳酸盐平衡进而造成海洋酸化(OA)。OA会影响海水和海洋污染物的理化性质,进而对生活在海洋表层的浮游藻类生理过程产生显著调控作用。海洋微藻作为海洋中主要的初级生产者,其生理功能与过程的正常进行对于海洋生态系统具有重要作用。本文综述了OA对海洋微藻光合固碳、钙化过程、固氮作用3个关键生理过程的调控作用和具体机制,总结了OA条件下,环境因素(如太阳辐射、温度、营养元素)对微藻生理过程和生长的影响,以及OA通过改变典型海洋污染物(如有机污染物、重金属、微塑料)的环境行为而对微藻生理过程的调控作用。最后,结合研究现状,对未来需要开展的研究方向进行展望。本文为进一步了解OA对海洋生态系统的潜在影响提供了重要信息。  相似文献

A continuous integrated bioprocess available from the earliest stages of process development allows for an easier, more efficient and faster development and characterization of an integrated process as well as production of small-scale drug candidates. The process presented in this article is a proof-of-concept of a continuous end-to-end monoclonal antibody production platform at a very small scale based on a 200 ml alternating tangential flow filtration perfusion bioreactor, integrated with the purification process with a model-based design and control. The downstream process, consisting of a periodic twin-column protein A capture, a virus inactivation, a CEX column and an AEX column, was compactly implemented in a single chromatography system, with a purification time of less than 4 hr. Monoclonal antibodies were produced for 17 days in a high cell density perfusion culture of CHO cells with titers up to 1.0 mg/ml. A digital twin of the downstream process was created by modelling all the chromatography steps. These models were used for real-time decision making by the implementation of control strategies to automatize and optimize the operation of the process. A consistent glycosylation pattern of the purified product was ensured by the steady state operation of the process. Regarding the removal of impurities, at least a 4-log reduction in the HCP levels was achieved. The recovery yield was up to 60%, and a maximum productivity of 0.8 mg/ml/day of purified product was obtained.  相似文献

Media preparation for perfusion cell culture processes contributes significantly to operational costs and the footprint of continuous operations for therapeutic protein manufacturing. In this study, definitions are given for the use of a perfusion equivalent nutrient feed stream which, when used in combination with basal perfusion medium, supplements the culture with targeted compounds and increases the medium depth. Definitions to compare medium and feed depth are given in this article. Using a concentrated nutrient feed, a 1.8-fold medium consumption (MC) decrease and a 1.67-fold increase in volumetric productivity (PR) were achieved compared to the initial condition. Later, this strategy was used to push cell densities above 100 × 106 cells/ml while using a perfusion rate below 2 RV/day. In this example, MC was also decreased 1.8-fold compared to the initial condition, but due to the higher cell density, PR was increased 3.1-fold and to an average PR value of 1.36 g L−1 day−1 during a short stable phase, and versus 0.46 g L−1 day−1 in the initial condition. Overall, the performance improvements were aligned with the given definitions. This multiple feeding strategy can be applied to gain some flexibility during process development and also in a manufacturing set-up to enable better control on nutrient addition.  相似文献

In this study, an ion exchange resin-based downstream-processing concept for imine reductase (IRED)-catalyzed reactions was investigated. As a model reaction, 2-methylpyrroline was converted to its corresponding product (S)-2-methylpyrrolidine with >99% of conversion by the (S)-selective IRED from Paenibacillus elgii B69. Under optimized reaction conditions full conversion was achieved using a substrate concentration of 150 and 500 mmol/L of d -glucose. Seven commercially available cation- and anion-exchange resins were studied with respect to their ability to recover the product from the reaction solution. Without any pretreatment, cation-exchange resins Amberlite IR-120(H), IRN-150, Dowex Monosphere 650C, and Dowex Marathon MSC showed high recovery capacities (up to >90%). A 150-ml preparative scale reaction was performed yielding ~1 g hydrochloride salt product with >99% purity. Any further purification steps, for example, by column chromatography or recrystallization, were not required.  相似文献
